近年、越境汚染はヨーロッパでも、また東アジアでも特有な黄砂現象などの新たな問題として認識されており、その気象動態などは過去に実績のからある程度明らかにされてきた。しかしながら、大陸飛来の黄砂微小粒子による健康影響を含め、越境汚染による健康影響に関しては、ほとんど研究がなされてこなかった。特に近年、日本においては、黄砂早期飛来が全国的に観測され、その同時期にスギ花粉飛散のピークも度々出現している。本来、スギ花粉は人間の鼻腔や口まで侵入し、下気道への吸入は無いと考えられてきたが、本研究代表者(王)のこれまでの研究では、スギ花粉のアレルゲン含有微小粒子(Cry j 1とCry j 2)が1.1 ミクロン以下に存在して大気中により長時間浮遊していることを明らかにした。都市部において、越境汚染で修飾された黄砂が、スギ花粉と接触することで、スギ花粉も修飾され、アレルギーを増悪させる可能性も考えられる。黄砂早期飛来時の花粉症状悪化事例が報道されているが、黄砂との複合影響は全く研究されておらず、予測もできないのが現状である。従って、花粉飛散から人体の呼吸器官への吸収までの間に花粉自体の微小化影響、微小なアレルゲン含有粒子の高感度計測、さらに黄砂粒子と汚染化学種の沈着や物理的・化学的修飾・毒性増強、花粉症罹患への黄砂と汚染化学種の複合影響を評価する必要がある。
本研究では、東アジアでの特有な黄砂飛来する現象が早まるに伴い、都市部で排出される汚染化学種によるスギ花粉や黄砂の修飾に関する影響評価を行い、その動態解析手法を開発し、花粉アレルゲンと黄砂・汚染化学種による花粉症罹患への複合影響について工学的解析を主とした分野融合型研究で、影響指標を探索し、国内外における免疫学的研究による生体影響評価手法の改善、環境汚染・花粉アレルゲン情報化システムの構築にも貢献しようとしている。
本研究は、我が国において既に深刻化している、また海外においても人体健康に懸念されつつある大陸飛来の黄砂現象、スギ花粉症の原因物質であるアレルゲンに着目し、これまでの基礎研究(王、研究成果の発表状況を参照)から得られた都市部におけるスギ花粉形態変化に伴うアレルゲン含有粒子(Cry j 1とCry j 2)の放出知見に加え、花粉アレルゲン含有微小粒子の発生機構を調べ、黄砂や汚染化学種の沈着影響評価、化学的修飾によるタンパク質構造変化を解析する。その上、花粉アレルゲン含有粒子と黄砂・汚染化学種による複合粒子状物質の形成機構解析、花粉症発症への関連研究を行い、微小粒子状の花粉アレルゲン含有粒子の放出による大気汚染化学種(水溶性有機炭素成分)としての自然起源寄与率を推定し、花粉飛散期における複合的な大気汚染現象形成への関与メカニズムを把握、嗅・肺神経・細胞などに介在可能なアレルゲン含有粒子と環境汚染化学種との複合粒子の曝露経路を解明していく。本計画研究の進展により、新たな情報が得られ、よりよい曝露指標開発、国際環境協力計測技術や学際的な知見に資する研究成果も期待できる。
より着実な成果を得るため、本研究は、研究代表者責任の下で、連携研究者(3名)の研究チームで互いに協力し、教育・研究を融合して大学院生(数名)の補助を仰ぎながら、本学の既設装置や設備を最大限に活用して、基礎研究→基盤整備→評価研究→学際的連携→国際共同研究など、段階的に研究を遂行してきている。

It is estimated that 15~30 % of the Japanese citizen were suffered from the cedar pollinosis which also the prevalence reached about 16%. The increment of the airborne cedar pollen counts is a main nominated reason, while many other factors such as the urban environmental pollution and human lifestyle are pointed out as well. The cedar pollinosis is one of the typical allergic diseases caused by two main kinds of allergens so-called Cry j 1 and Cry j 2 which exist in Japanese cedar pollen1~2). It had been considered that the pollen in coarse particles about 30 mm only deposits in upper part of the respiratory of human nasal cavity and mouth, and there was originally no inhalation to a lower respiratory tract. However, the existence of fine particles containing the Japanese cedar pollen allergens (fine Cry j 1 particles) has been confirmed in our previous studies. It is suggested presently that these fine particles can also invade into a lower respiratory tract and develop pollen asthma. In addition, long rage transportation of Asian dust from East Asian continent, called yellow sand, was found during the pollen scattering season in Japan due to the global warming. Therefore, interaction between pollen and yellow sand (Kosa) should be concerned.
In this study, we try to investigate airborne behavior of Japanese cedar pollen estimated their source apportionment to the urban atmosphere of Saitama City with the analysis of wind direction and trajectories, determine respirable Cry j 1 concentrations using the ELISA and surface plasmon resonance (SPR) methods, and review the relevance between airborne pollen counts and meteorological factors such rainfall, wind speed and trajectory analysis. In order to identify the influence factors on release and transition mechanism of pollen allergen contents to fine particle sizes, eluted allergen Cry j 1 contents from pollen grain were analyzed when pollen contacted with simulated rain of different concentrations of mineral and secondary formed air pollutants.
To clarify translation mechanism of small-sized allergen contents of Japanese cedar pollen existing together with urban atmospheric air pollutants, we researched size distribution of allergen (Cry j 1) particles containing in suspended particulate matter (SPM) of urban atmosphere. In addition, organic carbon (OC) contribution ratios caused from fine Cry j 1 contents and airborne Japanese cedar pollen grains were estimated. Rainfall ionic contents, the pollen counts and morphology of pollen grains during rainfalls were measured and investigated, since fine allergen particles were increased by sunny days after rainfall. Moreover, influences of air pollutants and yellow sand on modification of Cry j 1 were evaluated from molecular interaction between Cry j 1 and Cry j 1 antibody and change in molecular weight of water soluble protein in the pollen grains.
